T-cell dysfunction in chronic lymphocytic leukemia from an epigenetic perspective

چکیده

Cellular immunotherapeutic approaches such as chimeric antigen receptor (CAR) T-cell therapy in chronic lymphocytic leukemia (CLL) thus far have not met the high expectations. Therefore it is essential to better understand molecular mechanisms of CLLinduced dysfunction. Even though a significant number studies are available on function and dysfunction CLL patients, none examine at epigenomic level. In non-malignant research, epigenomics widely employed define differentiation pathway into exhaustion. Additionally, metabolic restrictions tumor microenvironment that cause often mediated by epigenetic changes. With this review paper we argue understanding (dys)regulation T cells patients should be leveled knowledge currently neoplastic B themselves. This will permit complete how these immune cell interactions regulate T- B-cell function. Here relate cellular phenotypic characteristics CLL-induced regulation emerging from viral infection models. proposes framework for future Tcell dysfunction, help guide improvements utility autologous based therapies CLL.

The PD-1/PD-L1 axis contributes to T-cell dysfunction in chronic lymphocytic leukemia.

Chronic lymphocytic leukemia is marked by profound defects in T-cell function. Programmed death-1 is a receptor involved in tumor-mediated immunosuppression through binding of the PD-L1 ligand. Multiparametric flow cytometry and immunohistochemistry were used to study PD-1/PD-L1 expression. Functional assays were used to determine the involvement of the PD-1/PD-L1 axis in T-cell responses. PD-1...

CD4+ T Cells are Exhausted and Show Functional Defects in Chronic Lymphocytic Leukemia

Background: Chronic lymphocytic leukemia (CLL) is the most common adult leukemia in the western world. This health problem is caused due to the accumulation of mature B-lymphocytes in the peripheral blood and bone marrow. In the course of cancer, CD4+ T cells become “exhausted” and characterized with poor effector functions and the expression of multiple inhibitory receptors. Objective: To inve...

Defective immunoregulatory T-cell function in chronic lymphocytic leukemia.

Chronic lymphocytic leukemia (CIL) of B-cell origin results in the malignant proliferation of small immunoglobulinbearing lymphocytes. There is currently a controversy in the literature regarding both the ability of this leukemic population to differentiate into mature plasma cells. as well as the ability of apparently normal T cells from these patients to regulate allogeneic B-cell differentia...
